Principle of the assay: CTSL1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CTSL1 antibody and an CTSL1-HRP conjugate. The assay sample and buffer are incubated together with CTSL1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTSL1 concentration since CTSL1 from samples and CTSL1-HRP conjugate compete for the anti-CTSL1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CTSL1 from the sample, fewer sites are left to bind CTSL1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTSL1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a lysosomal cysteine proteinase that plays a major role in intracellular protein catabolism. Its substrates include collagen and elastin, as well as alpha-1 protease inhibitor, a major controlling element of neutrophil elastase activity. The encoded protein has been implicated in several pathologic processes, including myofibril necrosis in myopathies and in myocardial ischemia, and in the renal tubular response to proteinuria. This protein, which is a member of the peptidase C1 family, is a dimer composed of disulfide-linked heavy and light chains, both produced from a single protein precursor. Multiple alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Apr 2012]
