Chicken gizzard tropomyosin.
Contents
Mouse ascites fluid, 1.2% sodium acetate, 2mg BSA, with 0.01mg NaN3 as preservative.
Reconstitution
1.2% sodium acetate or neutral PBS. If 1ml of PBS is used, the antibody concentration will be 100ug/ml.
Preparation and Storage
At -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Background: The tropomyosins are a family of actin filament binding proteins. These proteins were first isolated from skeletal muscle, but later identified in many nonmuscle tissues. Tropomyosins are ubiquitous proteins of 35 to 45 kD associated with the actin filaments of myofibrils and stress fibers. Vertebrates have at least 4 different tropomyosin genes; in humans, they are named TPM1, TPM2, TPM3, and TPM4. Tropomyosins expressed as different isoforms in muscle and non-muscle cells.

NCBI Description

This gene is a member of the tropomyosin family of highly conserved, widely distributed actin-binding proteins involved in the contractile system of striated and smooth muscles and the cytoskeleton of non-muscle cells. Tropomyosin is composed of two alpha-helical chains arranged as a coiled-coil. It is polymerized end to end along the two grooves of actin filaments and provides stability to the filaments. The encoded protein is one type of alpha helical chain that forms the predominant tropomyosin of striated muscle, where it also functions in association with the troponin complex to regulate the calcium-dependent interaction of actin and myosin during muscle contraction. In smooth muscle and non-muscle cells, alternatively spliced transcript variants encoding a range of isoforms have been described. Mutations in this gene are associated with type 3 familial hypertrophic cardiomyopathy. [provided by RefSeq, Jul 2008]

Plays a central role, in association with the troponin complex, in the calcium dependent regulation of vertebrate striated muscle contraction. Smooth muscle contraction is regulated by interaction with caldesmon. In non-muscle cells is implicated in stabilizing cytoskeleton actin filaments. Defects in TPM3 are a cause of nemaline myopathy type 1 (NEM1). Three alternatively spliced isoforms have been described.
