Function
Releases the supercoiling and torsionaltension of DNA introduced during the DNAreplication and transcription by transientlycleaving and rejoining one strand of the DNAduplex. Introduces a single-strand break viatransesterification at a target site in duplexDNA. The scissile phosphodiester is attackedby the catalytic tyrosine of the enzyme,resulting in the formation of aDNA-(3'-phosphotyrosyl)-enzymeintermediate and the expulsion of a 5'-OHDNA strand. The free DNA strand then rotatesaround the intact phosphodiester bond on theopposing strand, thus removing DNAsupercoils. Finally, in the religation step, theDNA 5'-OH attacks the covalent intermediateto expel the active-site tyrosine and restorethe DNA phosphodiester backbone (Bysimilarity). Regulates the alternative splicingof tissue factor (F3) pre-mRNA in endothelialcells. Involved in the circadian transcription ofthe core circadian clock componentARNTL/BMAL1 by altering the chromatinstructure around the ROR response elements(ROREs) on the ARNTL/BMAL1 promoter.

Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 2 weeks. For long term storage store at -20 degree C in small aliquots to prevent freeze-thaw cycles.

NCBI Description

This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus altering the topology of DNA. This gene is localized to chromosome 20 and has pseudogenes which reside on chromosomes 1 and 22. [provided by RefSeq, Jul 2008]
