Androgens exhibit a wide range of effects on the development, maintenance and regulation of male phenotype and male reproductive physiology. The androgen receptor (AR) is a member of the steroid superfamily of ligand-dependent transcription factors. ARs bind the two biologically active androgens, testosterone (T) and dihydrotestosterone (DHT), with high and nearly identical affinities; however, the rates of association and dissociation of T are about three times more rapid than those of DHT. This difference has resulted in speculation as to whether these differences in binding kinetics could account for the different physiological effects of T and DHT. A striking feature of AR is its rapid degradation in the absence of ligand. It is now well established that androgen binding results in an at least six-fold increase in androgen stability and that ligand-induced stabilization of AR is highly androgen- specific.

NCBI Description

The androgen receptor gene is more than 90 kb long and codes for a protein that has 3 major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. The protein functions as a steroid-hormone activated transcription factor. Upon binding the hormone ligand, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and then stimulates transcription of androgen responsive genes. This gene contains 2 polymorphic trinucleotide repeat segments that encode polyglutamine and polyglycine tracts in the N-terminal transactivation domain of its protein. Expansion of the polyglutamine tract causes spinal bulbar muscular atrophy (Kennedy disease). Mutations in this gene are also associated with complete androgen insensitivity (CAIS). Two alternatively spliced variants encoding distinct isoforms have been described. [provided by RefSeq, Jul 2008]
