Inactive FYN is phosphorylated on its C-terminal tail within the catalytic domain. Following activation by PKA, the protein subsequently associates with PTK2/FAK1, allowing PTK2/FAK1 phosphorylation, activation and targeting to focal adhesions.

NCBI Description

This gene is a member of the protein-tyrosine kinase oncogene family. It encodes a membrane-associated tyrosine kinase that has been implicated in the control of cell growth. The protein associates with the p85 subunit of phosphatidylinositol 3-kinase and interacts with the fyn-binding protein. Alternatively spliced transcript variants encoding distinct isoforms exist. [provided by RefSeq, Jul 2008]

Uniprot Description

Fyn: a tyrosine kinase of the Src family. Implicated in the control of cell growth. Plays a role in the regulation of intracellular calcium levels. Required in brain development and mature brain function with important roles in the regulation of axon growth, axon guidance, and neurite extension. Blocks axon outgrowth and attraction induced by NTN1 by phosphorylating its receptor DDC. Associates with the p85 subunit of phosphatidylinositol 3-kinase and interacts with the fyn-binding protein. Three alternatively spliced isoforms have been described. Isoform 2 shows a greater ability to mobilize cytoplasmic calcium than isoform 1. Induced expression aids in cellular transformation and xenograft metastasis. In squamous cell carcinoma, Fyn transduces signals from EGFR and Src and is required for cell migration and invasiveness. Activity linked to migration in a murine melanoma model. Appears to block late stage development of neuroblastoma. Mouse knockout deficient in kindling response, a model for human epilepsy.

Product Notes

The SYN / FYN fyn (Catalog #AAA249414) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-SYN / FYN Antibody (clone 2A10) IHC-plus reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SYN / FYN can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC) Paraffin, Immunocytochemistry (ICC), Immunofluorescence (IF), Western Blot (WB), Flow Cytometry (FC/FACS), ELISA (EIA). ELISA (1:10000) Flow Cytometry (1:200 - 1:400) ICC (1:200 - 1:1000) IF (1:1000) IHC-P (1:50) WB (1:500 - 1:2000). Researchers should empirically determine the suitability of the SYN / FYN fyn for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SYN / FYN, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
