Protein Phosphatase PPM1G is a PP2C type serine/threonine phosphatase. Unlike other PP2C members, PPM1G contains a large acidic domain. Magnesium or manganese is required for its phosphorylation activity. PPM1G has been shown to physically associate with spliceosomes in vitro and is believed to play a role in the early stages of splicesome assembly. In mouse, PPM1G has been found to localize to the nucleus and negatively regulate cell growth which suggests a role in cell cycle progression.

The protein encoded by this gene is a member of the PP2C family of Ser/Thr protein phosphatases. PP2C family members are known to be negative regulators of cell stress response pathways.
