The antibody detects an 89kDa* protein corresponding to the molecular mass of the cleaved fragment of PARP induced by staurosporine treatment of Jurkat cells. The antibody can be used to identify caspase-3 activity during apoptosis.

Clone 194C1439 was generated from a synthetic peptide corresponding to amino acids around the 214/215-cleavage site in human PARP. This sequence is well conserved in rat and mouse PARP, but the antibody has not been tested for these species reactivities.

The poly (ADP-ribose) polymerase (PARP) is involved in cell recovery from DNA damage, such as methylation of N3-adenine, which activates the base excision repair process. PARP [Poly (ADP-ribose) polymerase] is a 116kDa nuclear chromatin-associated enzyme that is cleaved during apoptosis by caspase-3 into a 24kDa fragment containing the DNA binding domain and an 89kDa fragment containing the catalytic and automodification domains. The 24kDa-fragment irreversibly bind to DNA and may contribute to the irreversibility of apoptosis by blocking the access of DNA repair enzymes to DNA strand breaks.
