Mouse PAI-1 Monoclonal Antibody | anti-PAI-1 antibody
PAI-1, Human, mAb MA-33H1F7
For unit size 0.5 mg or larger: PBS only (concentration< 0.5mg/ml)
Application Use: For Western blotting, dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. For functional studies, dilutions have to be optimized in user's experimental setting.
The epitope of monoclonal antibody MA-33H1F7 is predominantly composed of three residues (Lys154/Glu130/Arg131), positioned virtually linearly in the three-dimensional structure. The epitope of the antibody does not cover the complete alpha-helix F and turn connecting alpha-helix F and beta-strand s3A, but is restricted to the hinge region between alpha-helix F and the main part of the PAI-1 molecule.
The monoclonal antibody MA-33H1F7 is a ˜switching antibody, capable of inducing a non-inhibitory substrate form of PAI-1. It was shown to inhibit PAI-1 in a dose dependent manner.

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2009]
Uniprot Description
SERPINE1: a secreted protein that acts as 'bait' for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Belongs to the serpin family. Interacts with VTN. Binds LRP1B; binding is followed by internalization and degradation. Plasma levels of PAI-1 and VCAM-1 together may be useful in predicting post-operative recurrence in patients with colorectal cancer.

The PAI-1, Human, mAb MA-33H1F7 reacts with Reactivity: Human. Cross reactivity: Mouse, Rabbit, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PAI-1 can be used in a range of immunoassay formats including, but not limited to, FS, Immunoassay (IA), Western Blot (WB). Application Notes: W: A non-reduced sample treatment and SDS-Page was used. The band size is 52 kDa (Ref.5). Application Use: For Western blotting, dilutions to be used depend on detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. For functional studies, dilutions have to be optimized in user's experimental setting. Researchers should empirically determine the suitability of the PAI-1 serpine1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PAI-1, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
