Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Msx2, also known as HOX-8, is a homeobox containing transcription factor which is a member of the MSH family of homeobox proteins. Homeobox genes (HOX genes), encode DNA binding proteins that recognize specific sequences and modulate transcriptional activity (1). Msx-2 is believed to play a role in regulating bone development, particularly in sutures of cranial bone (5). Msx-2 is also expressed in sites of skin appendage formation and it has been proposed that Msx-2 is involved in the growth of skin and skin appendages (6). Msx2 mRNA is found expressed at higher levels in immature tumors, including a yolk sac tumor, seminoma, and chroiocarcinoma, and also in various tumors of epithelial origin (2). Msx2 gene expression is frequently activated in carcinoma-derived cell lines. The gene is inactive in NIH3T3 cells but is transcriptionally activated after transformation by v-Ki-ras suggesting that the Msx2 may play a role in cell transformation (3). Msx2 defects are a cause of craniosynostosis type 2 (CRS2), a disease that is characterized by premature fusion of calvarial sutures that leads to abnormal skull shape (4).

Uniprot Description

Function: Acts as a transcriptional regulator in bone development. Represses the ALPL promoter activity and antogonizes the stimulatory effect of DLX5 on ALPL expression during osteoblast differentiation. Probable morphogenetic role. May play a role in limb-pattern formation. In osteoblasts, suppresses transcription driven by the osteocalcin FGF response element (OCFRE). Binds to the homeodomain-response element of the ALPL promoter
