Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile (40-50%) glycerol, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

O6-methylguanine-DNA methyltransferase (MGMT) is a ubiquitous 22kD DNA repair protein, involved in the repair of mutagenic, carcinogenic, and cytotoxic O6-alkylguanine adducts, including primary alkyl adducts, induced by the chloroethylating and methylating agents used in the treatment of cancers. MGMT transfers alkyl groups from the O6 position of guanine in DNA to a cysteine residue in its sequence, restoring the integrity of DNA. This transfer is irreversible and inactivates MGMT; a cell's ability to withstand damage is directly related to the number of MGMT molecules within the cell and the rate of MGMT synthesis. MGMT repair thus represents a major form of chemotherapeutic drug resistance. The importance of MGMT in determining clinical response, both to chloroethylnitrosourea chemotherapeutic agents and to carcinogenesis by methylating agents, has been described. MGMT expression is highly variable in normal and tumor tissues, and also varies between individual cancer cells within a tumor.

MGMT: Involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) in DNA. Repairs alkylated guanine in DNA by stoichiometrically transferring the alkyl group at the O-6 position to a cysteine residue in the enzyme. This is a suicide reaction: the enzyme is irreversibly inactivated. Belongs to the MGMT family.
