Suitable for use in Western Blot.
Dilution: Western Blot: 1ug/ml detects JAK1 on NIH-3T3 mouse embryonic fibroblast cell line, Daudi human Burkitt's lymphoma cell line, HeLa human cervical epithelial carcinoma cell line and LNCaP human prostate cancer cell line.
Immunogen
Recombinant protein corresponding to aa32-286 of human Jak1 expressed in E. coli (P23458).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Janus Kinase 1 (Jak1) belongs to a family of protein tyrosine kinases that couple to cytokine receptors and are activated by ligand binding to these receptors. Activation of Jak1 occurs via phosphorylation at two adjacent tyrosine residues, Y1022 and Y1023, within the kinase domain. Jaks activate members of the STAT family of transcription factors by phosphorylating critical tyrosine regulatory sites. Jak1 is required for the activation of STAT1 and STAT2 in response to IFN-alpha.

Uniprot Description

Function: Tyrosine kinase of the non-receptor type. Appears to be required in early development for specific cell migrations (epiboly), expression of homeobox protein goosecoid and formation of anterior structures. Ref.1

Catalytic activity: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.

Subcellular location: Endomembrane system; Peripheral membrane protein

Developmental stage: Present in the unfertilized egg through to the blastula stage where it is distributed uniformly. Levels drop rapidly at four hours development, remain very low until 10 hours, then gradually increase from 12 hours with a rapid increase at 48 hours. At 48 hours it is concentrated in the region of the gill arches. Also present in the adult. Ref.1

Domain: Possesses two phosphotransferase domains. The second one probably contains the catalytic domain, while the presence of slight differences suggest a different role for domain 1.
