Interleukin 34 (IL-34) is secreted as a homodimer consisting of 39kD monomers. It belongs to no known cytokine family. The mature chain contains one potential site for N-linked glycosylation. IL-34 is widely expressed, but it is most abundant in the spleen. The receptor for IL-34 is colony-stimulating factor 1 receptor (CSF-1R). Functionally, IL-34 stimulates monocyte viability.

Signaling via CSF1R and its downstream effectors stimulates phosphorylation of MAPK1/ERK2 AND MAPK3/ERK1. Belongs to the IL-34 family. 2 isoforms of the human protein are produced by alternative splicing.
