Flow Cytometry (FC/FACS)

(Fig-2: Cell surface flow analysis of human IL3-RA in PBMCs (lymphocytes gated) using 0.5 ug/10^6 cells of antibody (Clone: ABM25C1). Green represents isotype control; red represents anti-IL3 RA antibody. Goat anti-mouse PE conjugate was used as secondary antibody. (Cells were incubated with primary antibody for 45 min. then washed twice with PBS by centrifuging at 1100 rpm for 5 min, followed by 30 min incubation with conjugated secondary antibody. Data acquisition was done after washing twice with PBS as mentioned above).)

Interleukin-3 receptor subunit alpha (IL-3RA) or CD123 is a high-affinity receptor for interleukin-3 (IL-3), which along with the primary signaling common Beta (Beta c) subunit (CD131) forms a heterodimer, enabling cell signaling. IL-3RA chain is a glycoprotein of 360 amino acidic residues, composed by an extracellular domain of 287 amino acid residues, involving a predicted Ig-like domain, two Fn III domains, a transmembrane domain of 30 amino acid residues and by an intracellular domain of 53 residues. It is expressed on the majority of CD34+ hemopoietic progenitors and its expression is rapidly lost during erythroid and megakaryocytic differentiation, moderately decreased during monocytic differentiation and is sustained in the granulocytic lineage. IL-3RA is also expressed in a variety of hematological neoplasms including blastic plasmacytoid dendritic cell neoplasm (BPDCN), acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), chronic myeloid leukemia (CML), Hodgkin’s lymphoma and hairy cell leukemia.

NCBI Description

The protein encoded by this gene is an interleukin 3 specific subunit of a heterodimeric cytokine receptor. The receptor is comprised of a ligand specific alpha subunit and a signal transducing beta subunit shared by the receptors for interleukin 3 (IL3), colony stimulating factor 2 (CSF2/GM-CSF), and interleukin 5 (IL5). The binding of this protein to IL3 depends on the beta subunit. The beta subunit is activated by the ligand binding, and is required for the biological activities of IL3. This gene and the gene encoding the colony stimulating factor 2 receptor alpha chain (CSF2RA) form a cytokine receptor gene cluster in a X-Y pseudoautosomal region on chromosomes X or Y. Alternatively spliced transcript variants encoding distinct isoforms have been found. [provided by RefSeq, Jun 2012]
