Principle of the assay: AQP-9 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AQP-9 antibody and an AQP-9-HRP conjugate. The assay sample and buffer are incubated together with AQP-9-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AQP-9 concentration since AQP-9 from samples and AQP-9-HRP conjugate compete for the anti-AQP-9 antibody binding site. Since the number of sites is limited, as more sites are occupied by AQP-9 from the sample, fewer sites are left to bind AQP-9-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AQP-9 concentration in each sample is interpolated from this standard curve.

NCBI Description

The aquaporins are a family of water-selective membrane channels. The protein encoded by this gene allows passage of a wide variety of noncharged solutes. It stimulates urea transport and osmotic water permeability; there are contradicting reports about its role in providing glycerol permeability. The encoded protein may also play a role in specialized leukocyte functions such as immunological response and bactericidal activity. [provided by RefSeq, Jul 2008]

Uniprot Description

AQP9: Forms a channel with a broad specificity. Mediates passage of a wide variety of non-charged solutes including carbamides, polyols, purines, and pyrimidines in a phloretin- and mercury-sensitive manner, whereas amino acids, cyclic sugars, Na(+), K(+), Cl(-), and deprotonated monocarboxylates are excluded. Also permeable to urea and glycerol. Belongs to the MIP/aquaporin (TC 1.A.8) family.

Product Notes

The Goat AQP-9 aqp9 (Catalog #AAA737161) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA737161 ELISA Kit recognizes Goat AQP-9. It is sometimes possible for the material contained within the vial of "Aquaporin 9, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
