Biological Significance
IKB alpha is the major IKB protein member of IKB family that function to inhibit the NF-KB transcription factor. IKBs have an N-terminal regulatory domain, followed by six or more ankyrin repeats and a PEST domain near their C terminus. IKBalpha inhibits NF-KB by masking the nuclear localization signals (NLS) of NF-KB proteins and keeping them in an inactive state in the cytoplasm. In addition, IKBalpha blocks the ability of NF-KB transcription factors to bind to DNA, which is required for NF-KB's proper functioning.
