Application Notes
Suitable for use in Immunocytochemistry, Flow Cytometry and ELISA.
Dilution: Immunocytochemistry: 10ug/ml detects GLP-2R in HeLa cells. Cells were fixed with PBS containing 10% normal donkey serum, 0.1% Triton X-100 and 1% BSA. After blocking, cells were incubated with diluted G2040-39D followed by NL557-coupled anti-mouse IgGin the dark. Between each step cells were washed with PBS, BSA.
Flow Cytometry: 25ug/ml. 10ul will label 1-2.5x10e5 cells iin 200ul total reaction volume.

HEK293 cells transfected with human GLP2R (O95838).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

GLP2R is a member of the class II glucagon-secretin G protein-coupled receptor superfamily. It is expressed on enteroendocrine cells in the gut where it mediates the positive effects of GLP2 on villus height, crypt depth and intestinal energy absorbtion. After stimulating a cAMP response, lipid raft-dependent internalization of GLP2R results in rapid desensitization to GLP2. Extracellular portions of human GLP2R show approximately 80% amino acid identity with corresponding regions of mouse GLP2R.

NCBI Description

The GLP2 receptor (GLP2R) is a G protein-coupled receptor superfamily member closely related to the glucagon receptor ans GLP1 receptor. Glucagon-like peptide-2 (GLP2) is a 33-amino acid proglucagon-derived peptide produced by intestinal enteroendocrine cells. Like glucagon-like peptide-1 (GLP1) and glucagon itself, it is derived from the proglucagon peptide encoded by the GCG gene. GLP2 stimulates intestinal growth and upregulates villus height in the small intestine, concomitant with increased crypt cell proliferation and decreased enterocyte apoptosis. Moreover, GLP2 prevents intestinal hypoplasia resulting from total parenteral nutrition. GLP2R, a G protein-coupled receptor superfamily member is expressed in the gut and closely related to the glucagon receptor (GCGR) and the receptor for GLP1 (GLP1R). [provided by RefSeq]
