Store at 4 degree C for one month or at -20 degree C for longer. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Mouse anti Human ErbB3 antibody, clone SGP1 recognizes human Receptor tyrosine-protein kinase erbB-3, also known as Proto-oncogene-like protein c-ErbB-3 or Tyrosine kinase-type cell surface receptor HER3. ErbB3 is a 1342 amino acid ~180 kDa glycoprotein containing a single protein kinase domain. Five isoforms of Erb-B3 are generated by alternative splicing. Mouse anti Human ErbB3 antibody, clone SGP1 recognizes a conformation dependent epitope on the extracellular protein core of ErbB3 rather than a carbohydrate moiety created during post-translational modification. ErbB3 is expressed widely in normal human tissues, but is over expressed in some tumours, including those of the breast, stomach, pancreas and colon (Lemoine et al. 1992). Mutations in the ERBB3 gene can lead to development of Lethal congenital contractural syndrome type 2, associated with atrophy of the spinal cord (Narkis et al. 2007).

NCBI Description

This gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. This membrane-bound protein has a neuregulin binding domain but not an active kinase domain. It therefore can bind this ligand but not convey the signal into the cell through protein phosphorylation. However, it does form heterodimers with other EGF receptor family members which do have kinase activity. Heterodimerization leads to the activation of pathways which lead to cell proliferation or differentiation. Amplification of this gene and/or overexpression of its protein have been reported in numerous cancers, including prostate, bladder, and breast tumors. Alternate transcriptional splice variants encoding different isoforms have been characterized. One isoform lacks the intermembrane region and is secreted outside the cell. This form acts to modulate the activity of the membrane-bound form. Additional splice variants have also been reported, but they have not been thoroughly characterized.
