Preparation and Storage

Western Blot (WB)

(The cell lysates (40ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human EpCAM antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.Lane 1.: MCF7 cell lysate Lane 2.: PC3 cell lysate Lane 3.: HeLa cell lysate)

Epithelial cell adhesion molecule (EPCAM) is a transmembrane glycoprotein mediating Ca2+-independent homotypic cell-cell adhesion in epithelia. EPCAM is also involved in cell signaling, migration, proliferation, and differentiation. Additionally, EPCAM has oncogenic potential via its capacity to upregulate c-myc, e-fabp, and cyclins A & E. Since EPCAM is expressed exclusively in epithelia and epithelial-derived neoplasms, EPCAM can be used as marker for various cancers. It appears to play a role in tumorigenesis and metastasis of carcinomas, so it can also act as a potential prognostic marker and as a potential target for immunotherapeutic strategies.

NCBI Description

This gene encodes a carcinoma-associated antigen and is a member of a family that includes at least two type I membrane proteins. This antigen is expressed on most normal epithelial cells and gastrointestinal carcinomas and functions as a homotypic calcium-independent cell adhesion molecule. The antigen is being used as a target for immunotherapy treatment of human carcinomas. Mutations in this gene result in congenital tufting enteropathy. [provided by RefSeq, Dec 2008]

EPCAM: a single-pass type I membrane protein of the EPCAM family. May act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium for providing immunological barrier as a first line of defense against mucosal infection. Expressed in almost all epithelial cell membranes but not on mesodermal or neural cell membranes. Found on the surface of adenocarcinomas including pancreatic carcinoma. May be a marker of cancer stem cells.
