Related Product Information for EPCAM elisa kit
Principle of the Assay: The Picokine™ Human EPCAM Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human EPCAM with a 96-well strip plate that is pre-coated with antibody specific for EPCAM. The detection antibody is a biotinylated antibody specific for EPCAM. The capture antibody is monoclonal antibody from mouse and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human EPCAM with immunogen: Expression system for standard: NS0; Immunogen sequence: Q24-K265. The kit is analytically validated with ready-to-use reagents. To measure Human EPCAM, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human EPCAM in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human EPCAM in the sample.

Background/Introduction: Epithelial cell adhesion molecule (EpCAM) is a transmembrane glycoprotein mediating Ca2+-independent homotypic cell-cell adhesion in epithelia. Also it is involved in cell signaling, migration, proliferation, and differentiation. Additionally, EpCAM has oncogenic potential via its capacity to upregulate c-myc, e-fabp, and cyclins A & E. Since it is expressed exclusively in epithelia and epithelial-derived neoplasms, EpCAM can be used as test marker for various cancers. It appears to play a role in tumorigenesis and metastasis of carcinomas, so it can also act as a potential prognostic marker and as a potential target for immunotherapeutic strategies.

NCBI Description

This gene encodes a carcinoma-associated antigen and is a member of a family that includes at least two type I membrane proteins. This antigen is expressed on most normal epithelial cells and gastrointestinal carcinomas and functions as a homotypic calcium-independent cell adhesion molecule. The antigen is being used as a target for immunotherapy treatment of human carcinomas. Mutations in this gene result in congenital tufting enteropathy. [provided by RefSeq, Dec 2008]

Uniprot Description

EPCAM: a single-pass type I membrane protein of the EPCAM family. May act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium for providing immunological barrier as a first line of defense against mucosal infection. Expressed in almost all epithelial cell membranes but not on mesodermal or neural cell membranes. Found on the surface of adenocarcinomas including pancreatic carcinoma. May be a marker of cancer stem cells.

Protein type: Membrane protein, integral; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 2p21

Cellular Component: apical plasma membrane; basolateral plasma membrane; cell surface; integral to plasma membrane; lateral plasma membrane; plasma membrane; tight junction

Molecular Function: protein binding; protein complex binding

Biological Process: negative regulation of apoptosis; positive regulation of cell proliferation; positive regulation of transcription from RNA polymerase II promoter; stem cell differentiation; ureteric bud development

Disease: Colorectal Cancer, Hereditary Nonpolyposis, Type 8; Diarrhea 5, With Tufting Enteropathy, Congenital

Product Notes

The Human EPCAM epcam (Catalog #AAA177637) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA177637 ELISA Kit recognizes Human EPCAM. It is sometimes possible for the material contained within the vial of "EPCAM PicoKine, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
