Purified by Protein G affinity chromatography.

Dilution: Flow Cytometry: Neat. 10ul labels 10e6 cells. Validated using human EDG1 transfected CHO-S cells. Cell surface expression is determined using 488nm wavelength excitation and monitoring emitted fluorescence with a detector optimized to collect peak emissions at 565-605nm.
Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. Add 10ul of E2208-01E to 1x10e6 cells and incubate for 30 minutes at RT.
3. Removed unbound antibody by washing the cells twice with Flow Cytometry Staining Buffer (F4568-10). Note that whole blood requires a RBC lysis step at this point.
4. Resuspend the cells in F4568-10 for analysis. As a control, cells in a separate tube should be treated with PE-labeled mouse IgG2b antibody.

It binds the ligand sphingosine-1-phosphate with high affinity and high specificity, and suggested to be involved in the processes that regulate the differentiation of endothelial cells. EDG-1: a G protein-coupled receptor for the lysosphingolipid sphingosine 1-phosphate (S1P). S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues. Highly expressed in endothelial cells and to a lesser extent, in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin. Suggested to be involved in the processes that regulate the migration/differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. S1P-induced endothelial cell migration requires AKT1-mediated phosphorylation of the third intracellular loop. Seems to be coupled to the G(i) subclass of heteromeric G proteins.
