Application Notes
Dilution: Flow Cytometry: 10ul labels 10e6 human EDG-1/S1P1-transfected CHO-S cells.

Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.
2. After blocking, 10ul of MBS602640 was added to up to 1x10e6 cells and incubated for 30 minutes at RT.
3. Unbound antibody was removed by washing the cells twice in Flow Cytometry Staining Buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.
4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2b antibody.

Other applications not tested. Optimal dilutions to be determined by the researcher.

NCBI Description

The protein encoded by this gene is structurally similar to G protein-coupled receptors and is highly expressed in endothelial cells. It binds the ligand sphingosine-1-phosphate with high affinity and high specificity, and suggested to be involved in the processes that regulate the differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. [provided by RefSeq]

EDG-1: a G protein-coupled receptor for the lysosphingolipid sphingosine 1-phosphate (S1P). S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues. Highly expressed in endothelial cells and to a lesser extent, in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin. Suggested to be involved in the processes that regulate the migration/differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. S1P-induced endothelial cell migration requires AKT1-mediated phosphorylation of the third intracellular loop. Seems to be coupled to the G(i) subclass of heteromeric G proteins.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "EDG1, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
