Immunogen
"Band 3" polypeptide of isolated desmosomes from bovine muzzle epidermis. Epitope: localized at the repeating unit domain ("RUD") of the cytoplasmic C-terminal part of the polypeptide.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. The desmosome is subdivided into two regions. The plaque region lies adjacent to the plasma, and is believed to contain molecules that attach the intermediate filament cytoskeleton to the desmosome. The core region is composed of transmembrane glycoproteins that are thought to mediate cell-cell adhesion. Desmogleins and desmocollins are the main desmosomal transmembrane proteins. These desmosomal glycoproteins belong to the members of the cadherin family of adhesion molecules. Three different isoforms of both desmogleins and desmocollins have been identified, named as desmoglein and desmocollin. Desmosomal cadherins showed differentiation-specific expression in the human epidermis, although the functional significance of this differential expression is not fully understood. Desmoglein-1 is synthesized exclusively in the suprabasal layers. The expression of desmoglein-2 varies in the basal layers, depending on the certain types of epidermis.

NCBI Description

Desmosomes are cell-cell junctions between epithelial, myocardial and certain other cell types. Desmoglein 1 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Currently, three desmoglein subfamily members have been identified and all are members of the cadherin cell adhesion molecule superfamily. These desmoglein gene family members are located in a cluster on chromosome 18. The protein encoded by this gene has been identified as the autoantigen of the autoimmune skin blistering disease pemphigus foliaceus. [provided by RefSeq]

DSG1: Component of intercellular desmosome junctions. Involved in the interaction of plaque proteins and intermediate filaments mediating cell-cell adhesion. Defects in DSG1 are the cause of palmoplantar keratoderma striate type 1 (SPPK1); also known as keratosis palmoplantaris striata I. SPPK1 is a dermatoligical disorder characterized by thickening of the skin on the palms and soles, and longitudinal hyperkeratotic lesions on the palms, running the length of each finger.
