Applicable Applications for anti-Dendritic Cell Immunoactivating Receptor antibody
ELISA (EL/EIA), Flow Cytometry (FC/FACS)
Application Notes
Flow Cytometry: This antibody was validated for flow cytometry using dendritic cells. Dilute this antibody to 25ug/ml and add 10 L of the diluted solution to 1-2.5 x 10e5 dells in a total reaction volume not exceeding 200 L. The binding of unlabeled monoclonal antibodies may be visualized by adding 10 L of a 25ug/ml stock solution of a secondary developing reagent such as goat anti-rat IgG conjugated to a fluorochrome., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse DCAR. The detection limit for rmDCAR is approximately 5 ng/well., Optimal dilutions should be determined by each laboratory for each application.
Immunogen
NS0-derived rmDCAR
Hybridoma
Produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a rat immunized with purified, NS0-derived, recombinant mouse Dendritic Cell immuno-Activating Receptor alpha isoform (rmDCAR; aa 42-209; Accession # AAP58453).
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for anti-Dendritic Cell Immunoactivating Receptor antibody
DCAR is a type II membrane protein belonging to the C-type lectin domain family and is designated CLEC4B. Two isoforms of DCAR exist, a 209 amino acid (aa) residue alpha isoform and a 176 aa form with a 33 aa deletion at the membrane proximal region of the extracellular domain. The DCAR extracellular domain contains a carbohydrate-recognition domain (CRD) that shares 91% amino acid sequence identity with the CRD of DCIR/CLEC4A. The DCAR intracellular domain is very short and lacks the ITIM motif found in DCIR. DCAR and DCIR are considered paired immunoregulatory receptors where DCAR activates through the ITAM of its associated adaptor molecule FcR.

Product Notes

The Dendritic Cell Immunoactivating Receptor (Catalog #AAA603769) is an Antibody produced from Rat and is intended for research purposes only. The product is available for immediate purchase. The Dendritic Cell Immunoactivating Receptor (DCAR, CLEC4B, C-type Lectin Domain Family 4 Member B) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Dendritic Cell Immunoactivating Receptor can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Flow Cytometry (FC/FACS). Researchers should empirically determine the suitability of the Dendritic Cell Immunoactivating Receptor for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Dendritic Cell Immunoactivating Receptor, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
