Broadly reacting cytokeratin antibody specific for cytokeratin 5. Stains basal cells in combined and stratified epithelial tissues. Recognizes the stem cell population, including the amplifying cell in the prostate epithelium. Species crossreactivity: human, rat, rabbit, hamster, canine, chicken, quail and porcine.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Cytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in epithelial tissues by at least 20 different polypeptides. They range in molecular weight from between 40kD and 68kD and isoelectric pH between 4.9-7.8. The individual cytokeratin polypeptides are designated 1-20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies.

K8: a type II cytoskeletal keratin. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. Phosphorylation of keratins at specific sites affects their organization, assembly dynamics, and their interaction with signaling molecules. Phsophorylated by p38 kinase, regulating cellular keratin filament reorganization. Phosphorylation on serine residues is enhanced during EGF stimulation and mitosis. Mutation of this protein is a risk factor for cryptogenic liver failure.
