Suitable for use in Direct ELISA, Immunohistochemistry and Western Blot.
Dilution: Direct ELISA: 0.5-1ug/ml. The detection limit for recombinant mouse Cyr61 is ~5ng/well.
Western Blot: 2ug/ml
Immunohistochemistry: 25ug/ml detects Cyr61 in frozen mouse trigeminal ganglia tissue sections.

Recombinant corresponding to aa176-281 of mouse Cyr61 expressed in E. coli (P18406).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Cyr61, also known as IGFBP-10 and CCN1, is a 50kD secreted matrix- and cell-associated glycoprotein that regulates the growth and adhesion of vascular endothelial cells, fibroblasts, and monocytes. Cyr61 interacts with cells that express integrins aVB3, aVB5, aMB2, and a6B1. Cyr61 is cleaved by plasmin within its VWF domain which generates an N-terminal fragment that is not associated with the matrix but retains the ability to induce endothelial cell migration. Cyr61 induces VEGF upregulation, angiogenesis, and tumorigenesis. Between amino acids 176-281, mouse Cyr61 shares 87% and 97% aa sequence identity with human and rat Cyr61, respectively.

NCBI Description

CYR61 is a secreted, cysteine-rich, heparin-binding protein encoded by a growth factor-inducible immediate-early gene. Acting as an extracellular, matrix-associated signaling molecule, CYR61 promotes the adhesion of endothelial cells through interaction with integrin and augments growth factor-induced DNA synthesis in the same cell type.[supplied by OMIM]

CYR61: Promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion. Appears to play a role in wound healing by up- regulating, in skin fibroblasts, the expression of a number of genes involved in angiogenesis, inflammation and matrix remodeling including VEGA-A, VEGA-C, MMP1, MMP3, TIMP1, uPA, PAI-1 and integrins alpha-3 and alpha-5. CYR61-mediated gene regulation is dependent on heparin-binding. Down-regulates the expression of alpha-1 and alpha-2 subunits of collagen type-1. Promotes cell adhesion and adhesive signaling through integrin alpha-6/beta-1, cell migration through integrin alpha-v/beta-5 and cell proliferation through integrin alpha-v/beta-3. Belongs to the CCN family.
