Performance
Ficoll prepared human peripheral blood mononuclear cells were stimulated by incubating 1 day at 2 x 106 cells/ml in RPMI 10% FBS media including 5 mug/ml Phytohemagglutinin-P. Five x 105 cells per tube were then washed and pre incubated 10 minutes with 20 mul human IgG at 250 mug/ml (to block non specific binding) after which they were incubated 45 minutes on ice with 80 mul of anti-CD94/Biotin at 5 mug/ml. Cells were washed twice and incubated with 2 degree reagent Streptavidin/R-Phycoerythrin, after which they were washed three times, fixed and analyzed by FACS. A net 20% sub population of the cells stained positive with a mean shift of 1.25 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 mul of 0.5 mg/ml anti-CD94 antibody.

Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.

Information: Human CD94 expression is restricted to natural killer (NK) cells and a small subset of T lymphocytes. CD94 is a type II membrane glycoprotein of the c-type lectin superfamily and essentially lacks a cytoplasmic tail. Antibody HP-3D9 recognizes the CD94 molecule of 70 kd. Antibody HP-3D9 inhibits IL-2 dependent proliferation of NK cells.

NCBI Description

Natural killer (NK) cells are a distinct lineage of lymphocytes that mediate cytotoxic activity and secrete cytokines upon immune stimulation. Several genes of the C-type lectin superfamily, including members of the NKG2 family, are expressed by NK cells and may be involved in the regulation of NK cell function. KLRD1 (CD94) is an antigen preferentially expressed on NK cells and is classified as a type II membrane protein because it has an external C terminus. Three transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
