Related Product Information for anti-CD8b antibody
The 341 monoclonal antibody reacts with rat CD8beta. CD8beta is a member of the immunoglobulin superfamily which heterodimerizes with CD8 alpha to form the CD8 antigen expressed on CD4+CD8+, double-positive thymocytes, the majority of MHC class I-restricted peripheral T cells, and on some macrophages. CD8beta expression is absent in athymic mice, which are populated by T-cells expressing CD8 alpha homodimers. This has led to the idea that CD8beta expression is dependent on the development of T cells in the thymus. CD8 binds to MHC class I expressed on the surface of antigen-presenting cells during antigen presentation, and participates in T-cell receptor signal transduction through association with the kinase Lck.

NCBI Description

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigens displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 beta chain isoforms. Multiple alternatively spliced transcript variants encoding distinct membrane associated or secreted isoforms have been described. A pseudogene, also located on chromosome 2, has been identified. [provided by RefSeq, May 2010]

Uniprot Description

Integral membrane glycoprotein that plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. In T-cells, functions primarily as a coreceptor for MHC class I molecule:peptide complex. The antigens presented by class I peptides are derived from cytosolic proteins while class II derived from extracellular proteins. Interacts simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen presenting cells (APCs). In turn, recruits the Src kinase LCK to the vicinity of the TCR-CD3 complex. A palmitoylation site in the cytoplasmic tail of CD8B chain contributes to partitioning of CD8 into the plasma membrane lipid rafts where signaling proteins are enriched. Once LCK recruited, it initiates different intracellular signaling pathways by phosphorylating various substrates ultimately leading to lymphokine production, motility, adhesion and activation of cytotoxic T-lymphocytes (CTLs). Additionally, plays a critical role in thymic selection of CD8+ T-cells.

Product Notes

The CD8b cd8b (Catalog #AAA5309346) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The CD8b antibody (PE) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD8b can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS). FC/FACS: = 0.125 ug/test A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 ul. Cell number should be determined empirically but can range from 105 to 108 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Researchers should empirically determine the suitability of the CD8b cd8b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD8b, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
