Performance
Five x 105 cultured Raji cells were washed and incubated 45 minutes on ice with 80 mul of anti-CD74/Biotin at a concentration of 5 mug/ml. Cells were washed twice and incubated with 2 degree reagent Streptavidin/R-Phycoerythrin, after which they were washed three times, fixed and analyzed by FACs. Cells stained positive with a mean shift of 1.77 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 inutes with 20 mul of 0.5 mg/ml anti-CD74 antibody.

Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.

50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Information: Human CD74 is expressed intracellularly and on the cell surface of MHC Class II positive cells such as B cells, monocytes, macrophages, Langerhans cells and dendritic cells. CD74 has several important functions relating to antigen presentation by MHC Class II molecules. Antibody M-B741 recognizes the CD74 molecule of about 33 kd.

NCBI Description

The protein encoded by this gene associates with class II major histocompatibility complex (MHC) and is an important chaperone that regulates antigen presentation for immune response. It also serves as cell surface receptor for the cytokine macrophage migration inhibitory factor (MIF) which, when bound to the encoded protein, initiates survival pathways and cell proliferation. This protein also interacts with amyloid precursor protein (APP) and suppresses the production of amyloid beta (Abeta). Multiple alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]

CD74: Plays a critical role in MHC class II antigen processing by stabilizing peptide-free class II alpha/beta heterodimers in a complex soon after their synthesis and directing transport of the complex from the endoplasmic reticulum to the endosomal/lysosomal system where the antigen processing and binding of antigenic peptides to MHC class II takes place. Serves as cell surface receptor for the cytokine MIF. A chromosomal aberration involving CD74 is found in a non-small cell lung tumor. Results in the formation of a CD74- ROS1 chimeric protein. 3 isoforms of the human protein are produced by alternative splicing.
