Application Notes
FLOW CYTOMETRY ANALYSIS:
Method:
1. Prepare a cell suspension in media A. For cell preparations, deplete the red blood cell population with Lympholyte®-Rat cell separation medium.
2. Wash 2 times.
3. Resuspend the cells to a concentration of 2x107 cells/ml in media A. Add 50 ul of this suspension to each tube (each tube will then contain 1x106 cells, representing 1 test).
4. To each tube, add 0.2 ug* of MBS520170.
5. Vortex the tubes to ensure thorough mixing of antibody and cells.
6. Incubate the tubes for 30 minutes at 4°C.
7. Wash 2 times at 4°C.
8. Add 100 ul of secondary antibody (Streptavidin-FITC) at a 1:500 dilution.
9. Incubate tubes at 4°C for 30 - 60 minutes (It is recommended that tubes are protected from light since most fluorochromes are light sensitive).
10. Wash 2 times at 4°C.
11. Resuspend the cell pellet in 50 ul ice cold media B.
12. Transfer to suitable tubes for flow cytometric analysis containing 15 ul of propidium iodide at 0.5 mg/ml in PBS. This stains dead cells by intercalating in DNA.

Media:
A. Phosphate buffered saline (pH 7.2) + 5% normal serum of host species + sodium azide (100 ul of 2M sodium azide in 100 mls).
B. Phosphate buffered saline (pH 7.2) + 0.5% Bovine serum albumin + sodium azide (100 ul of 2M sodium azide in 100 mls).
Immunogen
Rat T cell blasts (stimulated purified T helper cells with allogeneic irradiated rat spleen cells)
Donor
BALB/c spleen
Fusion Partner
Myeloma cell line NSO/1
Presentation
Biotin conjugated Ig buffered in PBS, 0.02% NaN3 and EIA grade BSA as a stabilizing protein to bring total protein concentration to 4-5 mg/ml.
Preparation and Storage
Store at 4 degree C. For long term storage, aliquot and freeze unused portion at -20 degree C in volumes appropriate for single usage. Avoid freeze/thaw cycles.

Related Product Information for anti-CD48 antibody
This anti-rat CD45 (Blast-1) monoclonal antibody recognizes a rat cell surface glycoprotein of 45 kDa that is present on a wide variety of hematopoietic cells and on endothelial cells. The antigen is identical to the mouse BCM1 antigen. This antibody inhibits allogeneic mixed lymphocyte reactions using lymph node cells as responders and spleen cells as stimulators. CD48 has recently been identified as a ligand of the NK cell inhibitory receptor CD244. This antibody is suitable for flow cytometry.

NCBI Description

This gene encodes a member of the CD2 subfamily of immunoglobulin-like receptors which includes SLAM (signaling lymphocyte activation molecules) proteins. The encoded protein is found on the surface of lymphocytes and other immune cells, dendritic cells and endothelial cells, and participates in activation and differentiation pathways in these cells. The encoded protein does not have a transmembrane domain, however, but is held at the cell surface by a GPI anchor via a C-terminal domain which maybe cleaved to yield a soluble form of the receptor. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Dec 2011]

Uniprot Description

Function: Ligand for CD2. Might facilitate interaction between activated lymphocytes. Probably involved in regulating T-cell activation.

Subunit structure: Interacts with CD244 in an heterophilic manner

By similarity.

Subcellular location: Cell membrane; Lipid-anchor › GPI-anchor.

Sequence similarities: Contains 2 Ig-like C2-type (immunoglobulin-like) domains.Contains 1 Ig-like V-type (immunoglobulin-like) domain.

Product Notes

The CD48 cd48 (Catalog #AAA520170) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Anti-Rat CD48 Biotin (Clone OX-45) (mouse IgG1) reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's CD48 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC). FLOW CYTOMETRY ANALYSIS: Method: 1. Prepare a cell suspension in media A. For cell preparations, deplete the red blood cell population with Lympholyte®-Rat cell separation medium. 2. Wash 2 times. 3. Resuspend the cells to a concentration of 2x107 cells/ml in media A. Researchers should empirically determine the suitability of the CD48 cd48 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD48, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
