Background: CCR4 (CC chemokine receptor 4) belongs to the rhodopsin family of G-protein-coupled receptors. Chemokine receptors are membrane-bound molecules composed of 7- transmembrane domains and are coupled to G-proteins (1,2). CCR4 binds the chemokines CCL17 (TARC) and CCL22 (MDC) and is highly expressed in most single-positive CD4(+) thymocytes especially in TH2- and regulatory T-cells (3,4). It plays a central role in T cell migration to the thymus, T cell maturation and education. CCR4 is often up-regulated in inflammation and cause conformational changes that trigger intracellular signaling pathways (2,5).

The protein encoded by this gene belongs to the G-protein-coupled receptor family . It is a receptor for the CC chemokine - MIP-1, RANTES, TARC and MCP-1. Chemokines are a group of small polypeptide, structurally related molecules that regulate cell trafficking of various types of leukocytes. The chemokines also play fundamental roles in the development, homeostasis, and function of the immune system, and they have effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis. CCR4: High affinity receptor for the C-C type chemokines CCL17/TARC and CCL22/MDC. The activity of this receptor is mediated by G(i) proteins which activate a phosphatidylinositol- calcium second messenger system. Can function as a chemoattractant homing receptor on circulating memory lymphocytes and as a coreceptor for some primary HIV-2 isolates. In the CNS, could mediate hippocampal-neuron survival. Belongs to the G-protein coupled receptor 1 family.
