Western Blot (WB) (Western blot analysis for detection of Caspase-3 activation in HeLa cells. Cells were treated with 2 mM staurosporine for different time periods. Caspase-3 activation is determined by cleavage of pro-Caspase-3, which generates 17 and 12kD, larger and smaller catalytic subunit, respectively.)

Specificity
Recognizes human caspase-3 protein. Detects both pro (~32kD) and the large subunit of the active/cleaved form (~14-21kD) of Caspase-3. The large subunit of the cleaved form may appear as one or two or even as a stack of bands depending on the presence or absence of the Caspase-3 pro-domain. Species Crossreactivity: mouse.

Supplied as a liquid in PBS, 0.02% sodium azide.
Applicable Applications for anti-Caspase 3, Active antibody
ELISA (EL/EIA), Western Blot (WB)
Application Notes
Suitable for use in Western Blot and ELISA.
Dilution: Western Blot: 2-3ug/ml
Optimal dilutions to be detemined by the researcher.
Immunogen
6xHis tag full-length human Caspase-3 protein, which was expressed in bacteria. Epitope: Recognizes an epitope in the large domain subunit of Caspase-3.
Positive Control
Staurosporine treated HeLa or jurkat cells.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Related Product Information for anti-Caspase 3, Active antibody
Caspases are a family of cysteine proteases that are key mediators of programmed cell death or apoptosis (1). The precursor form of all caspases is composed of a prodomain, and large and small catalytic subunits. The active forms of caspases are generated by several stimuli including ligand-receptor interactions, growth factor deprivation and inhibitors of cellular functions. All known caspases require cleavage adjacent to aspartates to liberate one large and one small subunit, which associate into a2b2 tetramer to form the active enzyme. Gene for Caspase-3 also known as Yama, CPP32, and apopain codes for a 32-kD protein (2-4). Caspase-3 cleaves the death substrate poly(ADP-ribose) polymerase (PARP) to a specific 85kD form observed during apoptosis and is inhibitable by the CrmA protein. Other Caspase-3 substrates include DNA-PK, actin, GAS2, and procaspase-6, etc. (5). Caspase-3 is activated by cleavage events at Asp-28/Ser-29 (between N-terminal pro-domain) and Asp-175/Ser-176 (between large and small subunits) to generate a large subunit of 17-kD and a small subunit of 12-kD (3).

Product Notes

The Caspase 3, Active (Catalog #AAA632143) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Caspase 3, Active (CPP-32, Apoptain, Yama, SCA-1) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Caspase 3, Active can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in Western Blot and ELISA. Dilution: Western Blot: 2-3ug/ml Optimal dilutions to be detemined by the researcher. Researchers should empirically determine the suitability of the Caspase 3, Active for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Caspase 3, Active, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
