Dilution: Western Blot: 1:1000 Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS,
0.1% Tween-20 at 4 degree C with gentle shaking, overnight.

Synthetic peptide surrounding Val184 of human CaMKII-A (KLH-coupled).
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

CaMKII is an important member of the calcium/calmodulin-activated protein kinase family, functioning in neural synaptic stimulation and T-cell receptor signaling (1,2). CaMKII has catalytic and regulatory domains. The binding of Ca2+/calmodulin to its regulatory domain releases its autoinhibitory effect and activates the kinase (3). The activated CaMKII further autophosphorylates at Thr286 to render the kinase constitutively active (3). The threonine phosphorylation state of CaMKII can be regulated through PP1/PKA. PP1 (protein phosphatase 1) dephosphorylates phospho-CaMKII at Thr286. PKA (protein kinase A) prevents this dephosphorylation through its inhibitory effect on PP1 (4).

Uniprot Description

Function: A key regulator of plasticity in synaptic physiology and behavior, alterations in its activity produce pleiotrophic effects that involve synaptic transmission and development as well as various aspects of behavior. Directly modulates eag potassium channels. Ref.6 Ref.7

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Enzyme regulation: CASK plays a role in regulation of CaMKII autophosphorylation. When complexed with CASK and in the presence Ca[2+]/CaM, autophosphorylation of Thr-287 causes constitutive activation of the kinase. In the absence of Ca[2+]/CaM, autophosphorylation of Thr-306 causes inactivation of the kinase. Ref.8

Subunit structure: Interacts with CASK. Ref.7

Tissue specificity: Expressed at a high level in the central nervous system during the late embryonic stage. In adults, expression is more abundant in the head than in the body. Ref.1 Ref.2

Post-translational modification: Autophosphorylation at Thr-287 is independent of autophosphorylation at Thr-306 and Thr-307.
