Testing Data
(Figure A: Hybridomas producing MAbs to Bim (peptide) were identified using clones of the IL-3-dependent mouse myeloid cell line FDC-P1 stably expressing Bcl-2 alone, or Bcl-2 plus mouse BimL mixed at a 1:1 ratio. Transfected cells were fixed, permeabilized and stained with the MBS602613, the mouse anti-Glu-Glu (EE) MAb (positive control) or with secondary antibody alone (negative control). Staining was visualised by FITC-conjugated goat anti-rat or anti-mouse IgG antibodies.)
Western Blot (WB)
(Figure B: Western blotting with anti-Bim MAb recognizes all known isforms of Bim. Western blotting of lysates of 293T cells cells transfected with EE-tagged forms of mouse or HA tagged forms of human Bim. Blots were probed with MBS602613, which detected EE mouse BimS, BimL, BimEL and EE human, BimSS, BimS, BimLS, BimL, BimELS, BimEL. Probing with an anti-Hsp-70 MAb was used as a loading control for all blots.)
Western Blot (WB)
(Figure C: Anti-Bim MAb detects endogenous Bim expression in mouse tissues but not those from Bim knockout mice. Bim protein was revealed in lysates from mouse tissues (5 x 106/lane) by Western blotting withMBS602613 and detection by ECL. Probing with an anti-Hsp-70 MAb was used as a loading control.)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene belongs to the BCL-2 protein family. BCL-2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. The protein encoded by this gene contains a Bcl-2 homology domain 3 (BH3). It has been shown to interact with other members of the BCL-2 protein family and to act as an apoptotic activator. The expression of this gene can be induced by nerve growth factor (NGF), as well as by the forkhead transcription factor FKHR-L1, which suggests a role of this gene in neuronal and lymphocyte apoptosis. Transgenic studies of the mouse counterpart suggested that this gene functions as an essential initiator of apoptosis in thymocyte-negative selection. Several alternatively spliced transcript variants of this gene have been identified. [provided by RefSeq, Jun 2013]
Uniprot Description
BIM: a pro-apoptotic member of the BCL-2 protein family. Interacts with other members of the BCL-2 protein family, including BCL2, BCL2L1/BCL-X(L), and MCL1, and act as an apoptotic activator. Its expression can be induced by nerve growth factor (NGF), as well as by the forkhead transcription factor FKHR-L1, which suggests a role in neuronal and lymphocyte apoptosis. Transgenic studies in the mouse suggested that this protein functions as an essential initiator of the apoptosis in thymocyte-negative selection. Nineteen alternatively spliced transcript variants of this gene have been reported.

Suitable for use in ELISA, Flow Cytometry, Immunocytochemistry, Immunoprecipitation,, Western Blot. Dilution: Immunoprecipitation: 2ug/ml Western Blot: 1-5ug/ml Optimal dilutions to be determined by researcher. Researchers should empirically determine the suitability of the BCL2L11 bcl2l11 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "BimS, EL, L, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
