Western Blot (WB) (Western blot analysis of Bid expression in Jurkat cell lysate (MBS179430).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-BID monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for BID )

Related Product Information for anti-Bid antibody
The major proteolytic product p15 BID allows the release of cytochrome c (By similarity). Isoform 1, isoform 2 and isoform 4 induce ICE-like proteases and apoptosis. Isoform 3 does not induce apoptosis. Counters the protective effect of Bcl-2.

NCBI Description

This gene encodes a death agonist that heterodimerizes with either agonist BAX or antagonist BCL2. The encoded protein is a member of the BCL-2 family of cell death regulators. It is a mediator of mitochondrial damage induced by caspase-8 (CASP8); CASP8 cleaves this encoded protein, and the COOH-terminal part translocates to mitochondria where it triggers cytochrome c release. Multiple alternatively spliced transcript variants have been found, but the full-length nature of some variants has not been defined. [provided by RefSeq, Jul 2008]

Uniprot Description

The major proteolytic product p15 BID allows the release of cytochrome c (). Isoform 1, isoform 2 and isoform 4 induce ICE-like proteases and apoptosis. Isoform 3 does not induce apoptosis. Counters the protective effect of Bcl-2.

Product Notes

The Bid bid (Catalog #AAA179430) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Bid Rabbit Monoclonal Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Bid can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunoprecipitation (IP), Immunofluorescence (IF), Immunohistochemistry (IHC), Immunocytochemistry (ICC), Western Blot (WB). WB: 1:500-1:1000 IHC: 1:50-1:200 ICC/IF: 1:50-1:200 IP: 1:10 FC/FACS: 1:50. Researchers should empirically determine the suitability of the Bid bid for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Bid, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
