It is recommended that the antibody be carefully titrated for optimal /formance in the assay of interest.
Biological Significance
The 15B8 monoclonal antibody reacts with human and mouse beta-catenin, one member of a family of catenins, which are intracellular proteins that interact with cadherins to mediate cellular adhesion. More specifically, beta-catenin binds to the cytoplasmic tail of E-cadherin. In addition, this molecule is a component of the canonical Wnt signaling pathway. In the absence of Wnt binding its receptor, beta-catenin is phosphorylated and resides in the cytoplasm where it is eventually targeted for degradation by ubiquitination. Upon Wnt binding, beta-catenin becomes dephosphorylated, translocates to the nucleus, and modulates gene expression in partnership with the transcription factors T cell factor and lymphocyte enhancer binding factor. Expression of beta-catenin is found in a wide variety of non-immune and immune tissues, including thymocytes and T and B lymphocytes. The Wnt beta-catenin signaling pathway has been demonstrated to play a crucial role in the development of T, B, and hematopoietic stem cells.

NCBI Description

The protein encoded by this gene is part of a complex of proteins that constitute adherens junctions (AJs). AJs are necessary for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. The encoded protein also anchors the actin cytoskeleton and may be responsible for transmitting the contact inhibition signal that causes cells to stop dividing once the epithelial sheet is complete. Finally, this protein binds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this gene are a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer. Three transcript variants encoding the same protein have been found for this gene.[provided by RefSeq, Oct 2009]

Uniprot Description

CTNNB1: a regulator of cell adhesion and a key downstream effector in the Wnt signaling pathway. Implicated early embryonic development and tumorigenesis. Phosphorylated and destabilized by CK1 and GSK-3beta. Stabilized cytoplasmic beta-catenin is a hallmark of a variety of cancers. Stabilized beta-catenin translocates to the nucleus, where it acts as a transcriptional activator of T-cell factor (TCF)-regulated genes. Interacts with the PDZ domain of TAX1BP3, inhibiting its transcriptional activity. Two alternatively spliced human isoforms have been described.

Product Notes

The CTNNB1 ctnnb1 (Catalog #AAA838004) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The beta Catenin antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's beta Catenin can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunohistochemistry (IHC), Immunoprecipitation (IP). FC: <= 1 ug/test A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 ul. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal /formance in the assay of interest. Researchers should empirically determine the suitability of the CTNNB1 ctnnb1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "beta Catenin, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
