Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to MCS. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for MCS and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain MCS, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of MCS in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Uniprot Description

MOCOS: Sulfurates the molybdenum cofactor. Sulfation of molybdenum is essential for xanthine dehydrogenase (XDH) and aldehyde oxidase (ADO) enzymes in which molybdenum cofactor is liganded by 1 oxygen and 1 sulfur atom in active form. In vitro, the C-terminal domain is able to reduce N-hydroxylated prodrugs, such as benzamidoxime. Defects in MOCOS are the cause of xanthinuria type 2 (XU2). Xanthinuria is characterized by excretion of very large amounts of xanthine in the urine and a tendency to form xanthine stones. Uric acid is strikingly diminished in serum and urine. In addition, patient suffering of XU2 cannot metabolize allopurinol into oxypurinol due to dual deficiency of xanthine dehydrogenase and aldehyde oxidase. Belongs to the class-V pyridoxal-phosphate-dependent aminotransferase family. MOCOS subfamily.

Product Notes

The Mouse Mocos mocos (Catalog #AAA288085) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA288085 ELISA Kit recognizes Mouse Mocos. It is sometimes possible for the material contained within the vial of "Molybdenum cofactor sulfurase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
