MANEAL Belongs to the glycosyl hydrolase 99 family.Glycoside hydrolases (also called glycosidases) catalyze the hydrolysis of the glycosidic linkage to release smaller sugars. They are extremely common enzymes with roles in nature including degradation of biomass such as cellulose and hemicellulose, in anti-bacterial defense strategies (e.g., lysozyme), in pathogenesis mechanisms (e.g., viral neuraminidases) and in normal cellular function (e.g., trimming mannosidases involved in N-linked glycoprotein biosynthesis). Together with glycosyltransferases, glycosidases form the major catalytic machinery for the synthesis and breakage of glycosidic bonds. Glycoside hydrolases are found in essentially all domains of life. In prokaryotes, they are found both as intracellular and extracellular enzymes that are largely involved in nutrient acquisition.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MANEAL in samples. An antibody specific for MANEAL has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMANEAL present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MANEAL is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MANEAL bound in the initial step. The color development is stopped and the intensity of the color is measured.
