Principle of the Assay||The Mouse MADCAM-1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse MADCAM-1 in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Mouse MADCAM-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and MADCAM-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse MADCAM-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MADCAM-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: MADCAM1 (Mucosal Vascular Addressin Cell Adhesion Molecule 1), also known as MACAM1, is a protein that is encoded by the MADCAM1 gene. By PCR-based analysis of somatic cell hybrids, Leung et al (1997) mapped the MACAM1 gene to chromosome 19. The protein encoded by this gene is an endothelil cell adhesion molecule that interacts preferentially with the leukocyte beta7 integrin LPAM-1 (alpha4/beta7), L-selectin, and VLA-4 (alpha4/beta1) on myeloid cells to direct leukocytes into mucosal and inflamed tissues. It is a member of the immunoglobulin superfamily and is similar to ICAM-1 and VCAM-1.

Uniprot Description

MADCAM1: Cell adhesion leukocyte receptor expressed by mucosal venules, helps to direct lymphocyte traffic into mucosal tissues including the Peyer patches and the intestinal lamina propria. It can bind both integrin alpha-4/beta-7 and L-selectin, regulating both the passage and retention of leukocytes. Isoform 2, lacking the mucin-like domain, may be specialized in supporting integrin alpha-4/beta-7-dependent adhesion strengthening, independent of L- selectin binding. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Motility/polarity/chemotaxis; Membrane protein, integral

Cellular Component: integral to membrane; membrane

Biological Process: cell adhesion; cell-matrix adhesion; heterotypic cell-cell adhesion; integrin-mediated signaling pathway; keratinocyte differentiation; leukocyte migration; leukocyte tethering or rolling; positive regulation of leukocyte migration

Product Notes

The Mouse MADCAM1 madcam1 (Catalog #AAA8244626) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244626 ELISA Kit recognizes Mouse MADCAM1. AAA Biotech's MADCAM-1 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the MADCAM1 madcam1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MADCAM-1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
