Principle of the Assay: The Picokine™ Mouse Madcam1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Mouse Madcam1 with a 96-well strip plate that is pre-coated with antibody specific for Madcam1. The detection antibody is a biotinylated antibody specific for Madcam1. The capture antibody is monoclonal antibody from rat, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Mouse Madcam1 with immunogen: Expression system for standard: NSO; Immunogen sequence: Q22-T365. The kit is analytically validated with ready to use reagents. To measure Mouse Madcam1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Mouse Madcam1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Madcam1 in the sample.

By PCR-based analysis of somatic cell hybrids, Leung et al(1997) mapped the MACAM1 gene to chromosome 19. The protein encoded by this gene is an endothelil cell adhesion molecule that interacts preferentially with the leukocyte beta7 integrin LPAM-1(alpha4 / beta7), Lselectin, and VLA-4(alpha4/beta1) on myeloid cells to direct leukocytes into mucosal and inflamed tissues. It is a member of the immunoglobulin superfamily and is similar to ICAM-1 and VCAM-1.

MADCAM1: Cell adhesion leukocyte receptor expressed by mucosal venules, helps to direct lymphocyte traffic into mucosal tissues including the Peyer patches and the intestinal lamina propria. It can bind both integrin alpha-4/beta-7 and L-selectin, regulating both the passage and retention of leukocytes. Isoform 2, lacking the mucin-like domain, may be specialized in supporting integrin alpha-4/beta-7-dependent adhesion strengthening, independent of L- selectin binding. 3 isoforms of the human protein are produced by alternative splicing.
