Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Leukosialin (SPN) ELISA Kit employs a two-site sandwich ELISA to quantitate SPN in samples. An antibody specific for SPN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Sialophorin (leukosialin) is a major sialoglycoprotein on the surface of human T lymphocytes, monocytes, granulocytes, and some B lymphocytes, which appears to be important for immune function and may be part of a physiologic ligand-receptor complex involved in T-cell activation.The nucleotide sequence of the 1.7-kb clone showed that the sialophorin transcript has a 3-prime-noncoding region of 587 nucleotides, is polyadenylated, and has the AATAAA polyadenylation signal. The derived sequence of 341 amino acids showed that the sialophorin protein has 3 domains: an extracellular domain of 195 amino acids, a hydrophobic transmembrane domain of 23 amino acids, and a C-terminal intracellular domain of 123 amino acids which contains a number of sites that might be phosphorylated by protein kinase C.

NCBI Description

This gene encodes a highly sialylated glycoprotein that functions in antigen-specific activation of T cells, and is found on the surface of thymocytes, T lymphocytes, monocytes, granulocytes, and some B lymphocytes. It contains a mucin-like extracellular domain, a transmembrane region and a carboxy-terminal intracellular region. The extracellular domain has a high proportion of serine and threonine residues, allowing extensive O-glycosylation, and has one potential N-glycosylation site, while the carboxy-terminal region has potential phosphorylation sites that may mediate transduction of activation signals. Different glycoforms of this protein have been described. In stimulated immune cells, proteolytic cleavage of the extracellular domain occurs in some cell types, releasing a soluble extracellular fragment. Defects in expression of this gene are associated with Wiskott-Aldrich syndrome. [provided by RefSeq, Sep 2017]

Is a counter-receptor for SN/Siglec-1. During T-cell activation is actively removed from the T-cell-APC (antigen-presenting cell) contact site thus suggesting a negative regulatory role in adaptive immune response. Interacts with HIPK2 via the cytoplasmic domain. Interacts with RDX. Cell surface of thymocytes, T-lymphocytes, neutrophils, plasma cells and myelomas.
