Principle of the Assay: LOX ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-LOX antibody and an LOX-HRP conjugate. The assay sample and buffer are incubated together with LOX-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the LOX concentration since LOX from samples and LOX-HRP conjugate compete for the anti-LOX antibody binding site. Since the number of sites is limited, as more sites are occupied by LOX from the sample, fewer sites are left to bind LOX-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The LOX concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is an extracellular copper enzyme that initiates the crosslinking of collagens and elastin. The enzyme catalyzes oxidative deamination of the epsilon-amino group in certain lysine and hydroxylysine residues of collagens and lysine residues of elastin. In addition to crosslinking extracellular matrix proteins, the encoded protein may have a role in tumor suppression. Defects in this gene are a cause of autosomal recessive cutis laxa type I (CL type I). Two transcript variants encoding different isoforms have been found for this gene.
