Scientific Background: KDM4C or lysine-specific demethylase 4C functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form (1). KDM4C is a member of the Jumonji domain 2 (JMJD2) family and contains one JmjC domain, one JmjN domain, two PHD-type zinc fingers, and two Tudor domains. KDM4C may contribute to tumor development and inhibition of KDM4C expression leads to decreased cell proliferation. KDM4C has been shown to play an important role in the development and/or progression of various types of cancer, including esophageal squamous cell carcinoma(2).

NCBI and Uniprot Product Information
NCBI Description
This gene is a member of the Jumonji domain 2 (JMJD2) family and encodes a protein with one JmjC domain, one JmjN domain, two PHD-type zinc fingers, and two Tudor domains. This nuclear protein functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form. Chromosomal aberrations and increased transcriptional expression of this gene are associated with esophageal squamous cell carcinoma. Alternative splicing results in multiple transcript variants. Uniprot Description
Function: Histone demethylase that specifically demethylates 'Lys-9' and 'Lys-36' residues of histone H3, thereby playing a central role in histone code. Does not demethylate histone H3 'Lys-4', H3 'Lys-27' nor H4 'Lys-20'. Demethylates trimethylated H3 'Lys-9' and H3 'Lys-36' residue, while it has no activity on mono- and dimethylated residues. Demethylation of Lys residue generates formaldehyde and succinate. Ref.6
Cofactor: Binds 1 Fe2+ ion per subunit. Ref.7
Subcellular location: Nucleus
By similarity.
Tissue specificity: Overexpressed in several esophageal squamous cell carcinomas (ESCs). Ref.1
Domain: The 2 Tudor domains recognize and bind methylated histones. Double Tudor domain has an interdigitated structure and the unusual fold is required for its ability to bind methylated histone tails
By similarity.
Sequence similarities: Belongs to the JHDM3 histone demethylase family.Contains 1 JmjC domain.Contains 1 JmjN domain.Contains 2 PHD-type zinc fingers.Contains 2 Tudor domains.
Sequence caution: The sequence BAA34500.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally shortened. The sequence CAI39608.1 differs from that shown. Reason: Erroneous gene model prediction.

Product Notes
The KDM4C kdm4c (Catalog #AAA515271) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 1-460 with tag GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant human KDM4C (1-460) was expressed by baculovirus in Sf9 insect cells. AAA Biotech's KDM4C can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the KDM4C kdm4c for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "KDM4C, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
