Biological Activity
Measured by its ability to induce IL-17 secretion by mouse splenocytes.
Preparation and Storage
Reconstitution: Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be sotred at 2-8 degree C. for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20 degree C to -80 degree C.

Storage/Stability: The lyophilized protein is stable at room temperature for up to 1 month. Working aliquots stored with a carrier protein are stable for at least 3 months at -20 degree C to -80 degree C. Avoid repeated freeze-thaw cycles.

IL-23 is a proinflammatory heterodimeric protein composed of two subunits, a unique p19 subunit and a p40 subunit, which is shared with IL-12. IL-23 is secreted by activated dendritic cells and macrophages, and signals though a receptor comprised of IL-23R complexed with IL-12Rbeta2. NCBI Description

This gene encodes a subunit of the heterodimeric cytokine interleukin 23 (IL23). IL23 is composed of this protein and the p40 subunit of interleukin 12 (IL12B). The receptor of IL23 is formed by the beta 1 subunit of IL12 (IL12RB1) and an IL23 specific subunit, IL23R. Both IL23 and IL12 can activate the transcription activator STAT4, and stimulate the production of interferon-gamma (IFNG). In contrast to IL12, which acts mainly on naive CD4(+) T cells, IL23 preferentially acts on memory CD4(+) T cells. [provided by RefSeq, Jul 2008]

Function: Associates with IL12B to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis. Ref.1 Ref.5 Ref.11

Subunit structure: Heterodimer with IL12B; disulfide-linked. The heterodimer is known as interleukin IL-23. Ref.14

Subcellular location: Secreted. Note: Secreted upon association with IL12B. Ref.1

Tissue specificity: Secreted by activated dendritic and phagocytic cells and keratinocytes. Also expressed by dermal Langerhans cells (at protein level). Ref.1 Ref.11

Developmental stage: Expressed by newborns dendritic cells. Ref.10

Induction: Up-regulated by a wide array of pathogens and pathogen-products together with self-signals for danger or injury. Up-regulated in psoriatic dermal tissues, in dendritic cells of multiple sclerosis patients and in tumors. Ref.6 Ref.7 Ref.8 Ref.9 Ref.11 Ref.12 Ref.13
