Insulin is a secreted peptide hormone that elicits metabolic effects such as increases in glucose uptake and glycogen synthesis leading to a decrease in blood glucose concentration. Insulin is first formed as a precursor molecule, preproinsulin, which is later cleaved to proinsulin and finally to the mature Insulin hormone. Mature Insulin consists of 51 amino acids, contained within an A chain and a B chain that are connected by 2 disulfide bridges. It increases cell permeability to monosaccharides, amino acids and fatty acids. Insulin is secreted by the pancreas at basal levels in the absence of exogenous stimuli, with secretion increasing in response to glucose. Insulin action is effected by the binding of Insulin to cell-surface receptors on the target cell membrane. Defects of Insulin are the cause of hyperproinsulinemia and of type-II diabetes mellitus.Insulin (4A2D11-H5) is also recommended for detection of Insulin in additional species, including bovine and porcine.

NCBI Description

After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into three peptides: the B chain and A chain peptides, which are covalently linked via two disulfide bonds to form insulin, and C-peptide. Binding of insulin to the insulin receptor (INSR) stimulates glucose uptake. A multitude of mutant alleles with phenotypic effects have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5' region and with the IGF2 gene at the 3' region. Alternative splicing results in multiple transcript variants.
