Application Notes
ELISA: Recombinant human Interleukin-1RA may be used as the standard for ELISA applications with either a purified human IL-1RA antibody or a biotinylated human IL-1RA antibody.
Western Blot: Recombinant huma Interleukin-1RA may be used as a positive control for Western Blotiing applications with either a purified human IL-1RA antibody or a biotinylated human IL-1RA antibody.
ELISA: Minimum Dilution: 0.2; Maximum Dilution: 0.4ng/well
Functional Assays: Minimum Dilution: 20; Maximum Dilution: 40ng/ml
Western Blotting: Minimum Dilution: 1.5; Maximum Dilution: 3.0ng/lane
Reconstitution
Reconstitute with 0.2ml distilled water. Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For extended storage, the addition of 0.1% bovine serum albumin (BSA) is recommended.

Preparation and Storage
Prior to reconstitution store at 4 degree C. Following reconstitution store at -20 degree C. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the protein. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 3 months from date of reconstitution

Interleukin-1 receptor antagonist (IL-1RA) is a naturally occurring protein which inhibits the activity of IL-1 alpha and IL-1 beta by competitively blocking binding to cell surface receptors for IL-1. In an assay of dose dependent inhibition of IL-1 stimulation of D10S cells, the ED50 was expected to be 20-40ng/ml.

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This protein inhibits the activities of interleukin 1, alpha (IL1A) and interleukin 1, beta (IL1B), and modulates a variety of interleukin 1 related immune and inflammatory responses. This gene and five other closely related cytokine genes form a gene cluster spanning approximately 400 kb on chromosome 2. A polymorphism of this gene is reported to be associated with increased risk of osteoporotic fractures and gastric cancer. Four alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

IL1RN: Inhibits the activity of interleukin-1 by binding to receptor IL1R1 and preventing its association with the coreceptor IL1RAP for signaling. Has no interleukin-1 like activity. Binds functional interleukin-1 receptor IL1R1 with greater affinity than decoy receptor IL1R2; however, the physiological relevance of the latter association is unsure. The intracellular form of IL1RN is predominantly expressed in epithelial cells. 4 isoforms of the human protein are produced by alternative splicing.
