Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate IL1RAPL1 in samples. An antibody specific for IL1RAPL1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL1RAPL1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL1RAPL1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL1RAPL1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 receptor family and is similar to the interleukin 1 accessory proteins. This protein has an N-terminal signal peptide, three extracellular immunoglobulin Ig-like domains, a transmembrane domain, an intracellular Toll/IL-1R domain, and a long C-terminal tail which interacts with multiple signalling molecules. This gene is located at a region on chromosome X that is associated with a non-syndromic form of X-linked intellectual disability. Deletions and mutations in this gene were found in patients with intellectual disability. This gene is expressed at a high level in post-natal brain structures involved in the hippocampal memory system, which suggests a specialized role in the physiological processes underlying memory and learning abilities, and plays a role in synapse formation and stabilization. IL1RAPL1: May regulate secretion and presynaptic differentiation through inhibition of the activity of N-type voltage-gated calcium channel. May activate the MAP kinase JNK. Plays a role in presynaptic and postsynaptic differentiation and dendritic spine formation in neurons. Defects in IL1RAPL1 are the cause of mental retardation X-linked type 21 (MRX21). Mental retardation is a mental disorder characterized by significantly sub-average general intellectual functioning associated with impairments in adaptative behavior and manifested during the developmental period. Non- syndromic mental retardation patients do not manifest other clinical signs. Belongs to the interleukin-1 receptor family. 1 isoforms of the human protein are produced by alternative splicing.
