The recombinant mouse IL13Ralpha2/Fc is a disulfide-linked homodimer after removal of the signal peptide. The reduced monomer consists of 561 amino acids and has a predicted molecular mass of 64.5 kDa. In SDS-PAGE under reducing conditions, the apparent molecular mass of rmIL13Ralpha2/Fc monomer is approximately 75-80 kDa due to glycosylation.

Background: Interleukin-13 receptor subunit alpha-2 (IL13RA2/IL-13RA2) is also known as also known as cluster of differentiation 213A2 (CD213A2), IL-13 receptor subunit alpha-2, IL-13R subunit alpha-2, and IL-13RA2. The IL13RA2 is often overexpressed in brain tumors, making Il13ra2 one of the vaccine targets for immunotherapy of glioma. IL13RA2/IL-13RA2 is a cancer-associated receptor that is present in greater than 80% of High Grade Astrocytomas (HGA) and has recently been recognized as a cytokine that predisposes breast cancer cells to metastasize. Expression of IL13Ralpha2 was rapidly lost from the surface of transduced cells grown in culture. The loss appeared to be related to ligands present in fetal bovine serum in the medium. None of the malignant glioma cell lines cultivated in vitro and tested to date exhibited the IL13Ralpha2 receptor. A recombinant virus (R5111) enters cells via its interaction with the IL13Ralpha2 receptor in a manner that cannot be differentiated from the interaction of wild-type virus with its receptors.

Description: A DNA sequence encoding the extracellular domain (Met 1-Lys 334) of mouse IL13Ralpha2 (NP_032382.1) was fused with the C-terminal polyhistidine-tagged Fc region of human IgG1 at the C-terminus.

NCBI Description

This gene encodes a receptor protein that binds to interleukin 13 (IL-13) with very high affinity. The encoded protein acts as a decoy receptor, and does not elicit any signal upon the binding of IL-13. Mice lacking the encoded protein exhibit increased levels of serum immunoglobulins, immune-dependent production of interferon gamma and, increased bone marrow macrophage progenitor frequency. Macrophages lacking the encoded protein release less nitric oxide and IL-12 in response to lipopolysaccharide. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Apr 2015]

IL13RA2: a type I membrane protein that binds with high affinity to interleukin-13 (IL13), but not to interleukin-4 (IL4). Lacks a cytoplasmic domain, and does not appear to function as a signal mediator. It is reported to play a role in the internalization of IL13. A member of the Cancer-Testis Antigen (CTA) superfamily. CTAs may play roles in embryonal development and tumor transformation or aspects of tumor progression. CTAs were once thought to be silenced in most normal adult tissues, with limited expression in fetal, placental, testis, and ovarian cells. These proteins are now known to be aberrantly expressed in various cancers and many are capable of eliciting humoral and cellular immune responses. The WSXWS motif appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding. The box 1 motif is required for JAK interaction and/or activation. Often overexpressed in brain tumors. A potential vaccine targets for immunotherapy of glioma.
