Principle of the Assay: The Picokine™ Human IL-12 (p70) Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human IL-12 (p70) with a 96-well strip plate that is pre-coated with antibody specific for IL-12 (p70). The detection antibody is a biotinylated antibody specific for IL-12 (p70). The capture antibody is monoclonal antibody from mouse and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human IL-12 (p70) with immunogen: Expression system for standard: NS0; Immunogen sequence: (I23-S328) + (R23-S219). The kit is analytically validated with ready-to-use reagents. To measure Human IL-12 (p70), add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human IL-12 (p70) in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human IL-12 (p70) in the sample.

NCBI Description

This gene encodes a subunit of a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. The cytokine is a disulfide-linked heterodimer composed of the 35-kD subunit encoded by this gene, and a 40-kD subunit that is a member of the cytokine receptor family. This cytokine is required for the T-cell-independent induction of interferon (IFN)-gamma, and is important for the differentiation of both Th1 and Th2 cells. The responses of lymphocytes to this cytokine are mediated by the activator of transcription protein STAT4. Nitric oxide synthase 2A (NOS2A/NOS2) is found to be required for the signaling process of this cytokine in innate immunity. [provided by RefSeq, Jul 2008]
