Intended Uses: This IL-36? ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat IL-36?. This ELISA kit for research use only!

Principle of the Assay: IL-36? ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IL-36? antibody and an IL-36? -HRP conjugate. The assay sample and buffer are incubated together with IL-36?-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-36? concentration since IL-36? from samples and IL-36? -HRP conjugate compete for the anti-IL-36? antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-36? from the sample, fewer sites are left to bind IL-36?-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-36? concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 cytokine family. Protein structure modeling indicated that this cytokine may contain a 12-stranded beta-trefoil structure that is conserved between IL1A (IL-A alpha) and IL1B (IL-1 beta). This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. Two alternatively spliced transcript variants encoding distinct isoforms have been reported. IL36B: Stimulates production of interleukin-6 and interleukin-8 in synovial fibrobasts, articular chondrocytes and mature adipocytes. Induces expression of a number of antimicrobial peptides including beta-defensins 4 and 103 as well as a number of matrix metalloproteases. Belongs to the IL-1 family. 2 isoforms of the human protein are produced by alternative splicing.
