Intended Uses: This sandwich kit is for the accurate quantitative detection of human Tyrosine Kinase 2 (also known as Tyk2) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.
Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human Tyk2 antibody. Tyk2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human Tyk2 Antibody is added and binds to Tyk2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated Tyk2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human Tyk2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The protein encoded by this gene belongs to the ubiquitin-specific proteases (UBP) family of enzymes that cleave ubiquitin from ubiquitinated protein substrates. It is highly expressed in liver and thymus, and is localized to the nucleus. This protein efficiently cleaves only ISG15 (a ubiquitin-like protein) fusions, and deletion of this gene in mice results in a massive increase of ISG15 conjugates in tissues, indicating that this protein is a major ISG15-specific protease. Mice lacking this gene are also hypersensitive to interferon, suggesting a function of this protein in downregulating interferon responses, independent of its isopeptidase activity towards ISG15.
